Thiosulfate Citrate Bile Salts Sucrose Agar is a selective medium for the isolation of pathogenic Vibrio. The formulation combines thiosulfate, citrate, ox bile and sodium taurocholate with sucrose, ferric citrate and the indicators bromothymol blue and thymol blue in an alkaline base (final pH 8.6±0.1).
Weigh 89 g and dissolve in 1000 mL of distilled water by boiling; cool to 45-50 °C and pour into sterile Petri dishes. Autoclaving is not required. Dehydrated powder (GF1070F) and granulated (KL1070F) forms are stored in a dark, dry place at room temperature and have a 3-year sealed shelf life.
